Alginate encapsulation of micro-cuttings in endangered Satureja khuzistanica species: a promising method for obtaining genetically stable plants with high rosmarinic acid content

Synthetic seed technology is a suitable approach for the rapid, and uniform mass production of Satureja khuzistanica Jamzad, valuable medicinal plant Lamiaceae family. In this study, encapsulation micro-cuttings S. using sodium alginate (SA) was investigated. order to determine best conditions encapsulation, different concentrations SA CaCl2·2H2O, types explants matrix culture media, growth regulators, cold storage synthetic seeds were tested. Also, genetic stability regenerated plants from inter simple sequence repeat (ISSR) molecular marker as well content rosmarinic acid in seed-derived evaluated. The use 3% 100 mM CaCl2·2H2O found be optimal gel complexation. Encapsulation nodal segments with 1/2 Murashige Skoog (MS) medium containing 2.5 µM 6-Benzylaminopurine resulted highest regrowth percentage (72.22%) rate (0.173). Pre-culture MS 5 indole-3-butyric (IBA) 0.2% activated charcoal 10 days IBA rooting (88.88%) plants. (61.11%) (0.173) obtained 2.0 thidiazuron (TDZ) liquid medium. placement germinated on coco peat substrate conversion seeds. encapsulated at 4 °C medium, compared without better (44.44%) (0.092) cold-stored occurred after 2 weeks. testing by ISSR showed that plantlets genetically similar their mother shoot tip-derived significantly enhanced up 7.77 times natural Genetic restoration programs, short-term storage, germplasm distribution could all benefit regeneration strategy reported here. By encapsulating alginate, we able produce stable high first time.
